19 Objective of the Course: To teach free-floating section technique and immunofluorescent staining used in histology laboratory.

21 Learning Outcomes:
1 Ability to use total perfusion technique.;
2 Learning the usage areas and technical details of free floating section technique.;
3 Learning the application of immunofluorescence staining technique on free floating sections.;
4 To learn using cryotome in histology laboratory.;
22 Course Content:
Week Theoretical Practical
1 Routine histology technique.
2 Use of a microtome.
3 Tissue detection, fixing solutions, solutions appropriate to cryo-section.
4 Total perfusion technique.
5 Usage areas of cryotome, reasons of preference.
6 Using cryotome, collecting free floating sections.
7 Preparation of Cryo protector solution and washing solutions.
8 Washing of tissues, selection of appropriate sections.
9 Selection of appropriate primary antibody and fluorescent labeled secondary antibodies.
10 Application of fluorescent staining 1 (primary antibody in well plate)
11 Application of fluorescent staining 2 (Use of fluorescently labeled antibodies)
12 Taking free floating sections on to microscope slides, use of fluorescence microscope, storage of sections.
13 Double immunofluorescent staining.
14 Free floating section technique in central nervous system.
